The Lions trailed 41-17 heading into the final two minutes but pounced on the Niners when they switched off.
They scored two TDs in less than a minute and had one final chance to force OT after Deebo Samuel fumbled with 58 seconds left.
Detroit failed to take advantage though and started the season with a loss.
In his Detroit debut Jared Goff went 38/57 for 338 yards, three touchdowns and an interception.
Running backs Jamaal Williams and De’Andre Swift combined for 93 yards rushing and one TD, as well as 121 yards receiving and a score.
TJ Hockenson led in receiving, catching eight of his 10 targets for 97 yards and a touchdown.

Green Bay’s Week 1 loss was a surprise but there is too much talent on this roster for that to become the norm.
